【摘要】目的:探讨Toll样受体4(tolllike receptor 4,TLR4)和CD14在晶状体上皮细胞(lens epithelial cell, LEC)的表达。方法:采用一步法反转录多聚酶链反应(polymerase chain reaction, PCR)检测LEC中TLR4 mRNA和CD14 mRNA的表达。结果:用RTPCR检测分别获得451bp和348bp的清晰扩增条带,经基因测序证实为TLR4 mRNA和CD14 mRNA。结论:晶状体上皮细胞可表达TLR4和CD14。

0 引言
Toll样受体(tolllike receptor,TLR)对机体免疫特别是感染性免疫具有十分重要的意义。迄今在人细胞膜上已发现11种TLR(TLR1 TLR11),其中TLR4是研究最多的一种Toll样受体蛋白。有关TLR4的研究被《Science Watch》杂志列为2001年十大生物科学重要进展之一[1]。TLR4能识别G外膜的主要成分脂多糖(haride, LPS),并介导LPS引起的炎症反应。LPS激活TLR4的信号途径需要CD14的参与,CD14能促进TLR4对LPS的应答。已有研究表明,TLR4在许多细胞均有表达[2]。但至今尚不知在晶状体上皮细胞(lens epithelial cell, LEC)是否表达TLR4,也不知LEC是否表达CD14。白内障是当今世界的主要致盲性眼病,超声乳化白内障囊外摘除联合人工晶状体植入手术是目前的主要复明手段。但研究表明术后眼内发生了炎症反应[35]。白内障术中及术后炎症介质释放、补体激活、血房水屏障破坏等均刺激LE
