【摘要】目的: 探讨阿司匹林对胃癌细胞SGC7901增殖及核因子κB p65(NFκB p65)表达的影响。方法: 培养胃癌细胞株SGC7901,采用MTT法检测阿司匹林在不同浓度和不同作用时间下对胃癌细胞增殖的影响;DNA琼脂糖凝胶电泳检测细胞凋亡;蛋白质印迹法和免疫荧光法检测阿司匹林对胃癌细胞 NFκB p65表达的影响。结果: MTT结果显示,~10 mmol/L浓度区间内,随浓度增加和时间的延长,阿司匹林对SGC7901细胞的抑制率逐渐增加。SGC7901细胞经10 mmol/L阿司匹林作用6 h和12 h后,细胞发生凋亡。随着阿司匹林作用浓度增加和作用时间延长,胃癌细胞 NFκB p65的表达逐渐降低。结论: 阿司匹林对胃癌细胞SGC7901的增殖具有抑制作用,且与阿司匹林的浓度和作用时间呈正相关,其作用机制可能与通过抑制胃癌细胞NFκB p65的表达而使细胞发生凋亡有关。

[Abstract] Objective: To investigate the effect of aspirin on proliferation and nuclear factor kappa B expression in gastric cancer cell line SGC
7901. Methods: Gastric cancer cell line SGC7901 was treated with aspirin of different concentrations or different time, effect of aspirin on proliferation and apoptosis in SGC7901 was evaluated by MTT assay and DNA agarose gel electrophoresis; expression of nuclear factor kappa B p65 was determined by Western blotting and immumofluorescence method. Results: Aspirin can inhibit the proliferation of SGC7901 among concentrations of ~10 mmol/L. As the dose and time increased, the inhibitory of aspirin on proliferation of SGC7901 was significantly increased. Apoptosis was detected by DNA agarose gel electrophoresis after treating with aspirin of 10 mmol/L for 6 or 12 hours. Aspirin reduced expression of NFκB p65 in a concentration and time dependent pattern. Conclusion: Aspirin inhibited proliferation of gastric cancer cell line SGC7901; the inhibition of NFκB p65 expression inducing cellular apoptosis may ascribed to mechanism of antiproliferation activity of aspirin.

近年来研究表明,阿司匹林对胃肠道肿瘤具有预防作用[
1],能抑制肿瘤细胞增殖,诱导肿瘤细胞凋亡,其作用机制包括多个方面但并不很明确。过去大量研究显示,阿司匹林通过抑制环加氧酶2(COX2)活性,下调COX2的表达而发挥抗肿瘤效应[2]。但目前很多研究提示,阿司匹林能通过非COX2依赖性途径发挥抗肿瘤作用。核因子κB(NFκB)信号通路便是其中之一。NFκB是细胞中的重要转录因子,在受到各种激活因子如肿瘤坏死因子(TNF)刺激时,NFκB与抑制分子IκB解离而被活化,p50p65二
