番茄中玉米素提取条件的优化#

摘要:结合高效液相色谱(HPLC)研究了番茄中玉米素的提取纯化方法。玉米素对光不敏
感,可在 60℃下浓缩。玉米素固相萃取时,C18 小柱用 2ml 80%甲醇活化后再用 1ml pH=3.
0 的乙酸水活化效果较好;从番茄中提取玉米素时,C18 小柱洗脱剂使用 45%甲醇除杂效果
较好。准备进行 HPLC 的样品是否用流动相溶解对检测结果没有影响。本实验为研究玉米素
对番茄再生的调控提供了有效的提取方法。

Optimization the method used in zeatin purification from
tomato

Abstract: The study was conducted to establish the methods which to purify zeatin in tomato.
Zeatin was detected by high performance liquid chromatography (HPLC). Zeatin was insensitive
to light,and stable at 60℃. C18 SPE column would be washed sequentially with 2ml 80%
methanol,1ml water–acetic acid (pH=) before applying the zeatin to the column. 45% ethanol
would be used as eluent while purifying the zeatin from tomato. There was no effect to result if the
sample was dissolved with the mobile phase of HPLC. The paper gave the methods of purifying
zeatin for studies on regulation of tomato regeneration by zeatin.
