摘要: 目的】初步探索葛根总黄酮对 HL-60 细胞可能的分化作用。方法】0~100μg/ml PRF
干预 HL-60 细胞 24、48 及 72 小时,MTT 法检测增殖抑制率。0~50μg/ml PRF 干预 HL-60
细胞 48 小时,瑞氏染色法观察细胞形态学改变,检测细胞 NBT 积分。流式细胞术检测细胞
表面分化抗原 CD11b、CD14、CD15、CD13 及 CD33 及细胞周期进程。【结果】10、30、
50 μg/ml PRF 可抑制 HL-60 细胞的增殖;部分细胞形态出现分化特征; G0-G1 期细胞比
例增加,S 期比例减少,影响细胞周期进程;细胞表面分化抗原 CD11b、CD15 表达增加,
CD14 低表达,CD33 表达下降,CD13 表达无明显变化;NBT 积分随药物浓度增加而升高。
【结论】较低浓度 PRF(0~50μg/ml)可体外抑制 HL-60 细胞增殖,影响细胞周期进程;诱
导 HL-60 细胞向粒细胞样细胞分化,具体机制有待进一步研究。
